A potential antimalarial prospect should have high efficacy with low toxicity. SynriamTM antimalarial drug has been proven to be safe and effective for the treatment of malaria in human with dearth information on rats. This study investigated its potential toxicity from prolonged administration on lipid profiles and haematological indices in rats. Thirty-five Wistar rats were randomized into five groups (A – E) of seven rats each. Animals in group A (control) received 0.5 mL distilled water, while groups B, C, D, and E respectively received 4.0, 8.0, 16.0 and 32.0 mg/kg bwt of the drug once daily for 28 days. The concentrations of serum TC, LDL and atherogenic index showed significant (p<0.05) decrease, while serum TRIG, HDL, and VLDL concentrations had no effect (p>0.05). The drug also had no effect (p>0.05) on the levels of the red blood cell count, PCV, Hb, MCV, MCH, and MCHC. Similarly, it had no effect (p>0.05) on the levels of the WBC, lymphocytes and neutrocytes, while the platelet counts had slight (p<0.05) decrease. Hence, SynriamTM is relatively safe at doses of 4.0-32.0 mg/kg bwt in rats when taken once daily with minimal effects on the lipid profiles and haematological indices.

The study was to determine the level of toxicity of repeated administration of ethanol extract of a medicinal mushroom, Daldinia concentrica in animal model. Wistar albino rats of both sexes were sorted randomly into four groups of five rats each. Graded doses of 200, 400 and 800 mg/kg of the extract were administered orally, once daily for eight consecutive days to the rats in the respective groups, while control group received 10% tween 80 in distilled water only. The animals were monitored daily for behavioural activities, while effects on body weight, organ-body weight indices and haematological parameters were evaluated at the end of the treatment period. There was significant (P< 0.05) and dose dependent increase in the body weight of the test groups compared with the control. The organ-body weight indices did not differ significantly (P>0.05), while monocyte, platelet and plateletcrit values were elevated among the 200 and 400 mg/kg body weight treatments. However, the observable changes were not sufficient to establish any toxic effect arising from the sub-acute oral administration of the crude ethanol extract of D. concentrica at all test doses in this study. Rather, the effects on body weight and haematological parameters suggested that the extract contains nutritional and immune boosting properties.

The effect of the ethanol extract of Aloe barbadensis root on selected serum biochemical parameters was investigated.Sixty male Wistar rats were randomly selected and administered 100, 200 and 400 mg/kg of the extract and 0.2ml/kg of distilled water (control) for 14 days. The results of the liver marker enzymes revealed that there were non-significant changes (p>0.05) in alkaline phosphatase (ALP), aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels on days 1, 7 and 14 of treatment. The concentrations of other liver biomarkers (total bilirubin, direct bilirubin, albumin and total proteins) were also not significantly (p>0.05) altered. Selectively, on day 7, the concentration of total protein was significantly (p<0.05) different at all treated doses from the control. The extract had no significant (p>0.05) effect generally on the kidney's functional indices. At 100 and 200 mg/kg, selective alterations in some renal biomarkers (creatinine, urea, phosphorous and chloride) on different days of treatment were noticed. The results indicate that within the doses used in the study, the ethanol extract of A. barbadensis root is relatively non-toxic with no significant localized toxicity
